What do you want your first mission to be?
I want my first mission to be an easy one, so I can get use to my crew and my ship. To get use to the game.

That kind of goes without saying.
I would like something based from Earth as though I've just graduated the Academy. Run the first few days at low rank and short range.

I'd settle for anything with my first mission.
I'd be too in awe at finally playing STO to be picky.
However,I'd like my first mission to be on a runabout, and I'd like for it to take place in a beautiful region of space that shows off how lovely the graphics engine really can be.

I was going to say a shakedown cruise of your vessel, make sure the warp core doesnt blow or shake apart, maybe answer a distress call on a disabled ship.

I think it will be some scripted even that happens during maybe a shakedown cruise (a timeless classic), or during normal small-ship operations.
My first real mission? Head out and explore... I'm sure we'll run into enough trouble :D

I would imagine the first mission/tutorial would involve some sort of event where your ships captain is incapacated and you take over, thus you become captain without years of service.

I think it should be something simple as you are starting out as a very junior officer, I assume. Maybe just finding your way around the ship/starbase/orbiting station, etc., would be a good place to start

It has to be a simulation, (perhaps the Kobiashi Maru?) to teach you the basics of starship command and control.

First mission will likely be a tutorial while you are in the academy. There will likely be a space and ground section.

"Mr Savok, have you ever piloted a starship out of space dock"
Seems like a good first mission

this is not what i would "want", but more what i am expecting...........
first mission will be a milk run. either cargo transport (medicine, machine parts, suppies, etc..) that need to go to some outpost. your "goods"are highjacked and u need to get them back.
or
first mission is a diplomatic currier mission. deliver diplomat to such and such destination. ur ship is attacked and the diplomat is killed. u are forced to complete the diplomatic mission w/o the diplomat.

I would want a shakedown cruise which doubles as a tutorial for the game. It should be quite easy in order to start to master the game mechanics and decision making processes.

Saavik :p Kirstie Alley was also the best Saavik to, just sucks she didn't want to be associated with sci fi.

I'd like to have a short academy tutorial before flying a runabout to my new ship.

Some sort of diplomatic/trade/dispatch mission to Vulcan. That's the first thing I want to do.
After that, it's all gravy.

Federation: A mission that starts out as a simple patrol of sector 001 but you receive a distress signal. you investigate and find a damaged federation Starship. It claims to have been attacked by the Klingons. suddenly a Klingon ship decloaks off your port and fires. You destroy it and report to Starfleet Command to claim your reward for saving the vessel. This could be the first part of the story which leads on to find that an alien race is capturing ships to use to make the federation and Klingons go to war, leaving them weak and easy to dominate.
Same with Klingons only with alternate starships

Im would think it would be a shakedown cruise, or if your character is not a captain yet, I've been pondering the last mission of the captain before you.

I think my first personal mission would be to take my shuttle and try to slingshot myself around the sun into the past... probably incinerating myself in the progress. So I guess my first real mission would be to get a new ship after I destroyed my other one.

The best kind of first mission is where everything is going right, then goes horribly wrong:D Like you are flying with a diplomat to a planet, and bam fleet of ships comes and starts to form up to attack! Captain is killed (somehow and the fact of how he dies does not matter to me, like an explosion kills him). Then you take control of the ship and depending on what you try to do, your character will be affected. You could try to communicate, attack or use the area of space to your advantage(thus running/guerrilla warfare). Then when you are done, this mission will set the beginnings of your Captain, and how the Federation feels about him. Maybe also directing you to use a different ship.

you should be thrown into a crazy situation as kinda a quick tutorial, then once the first mission is finished you graduate from the academy and bam you are in the game

I think a nice first mission could be simply as reporting to your ship :)
